薛向东,长聘教轨副教授,课题组组长,博士生导师。

博士毕业于中国科学院国家纳米科学中心,师从梁兴杰研究员。2015年至2020年,分别在美国华盛顿大学(Samuel Wickline课题组)和加州大学戴维斯分校(Yuanpei Li课题组)从事博士后研究工作。2020年12月以“高层次人才引进”的方式加入15vip太阳集团,长期致力于:纳米药物制剂的开发及其对癌症等疾病的精准诊疗。目前已发表高水平SCI论文60余篇,论文被引4000余次,H因子31。包括以第一或通讯作者在 Nature Nanotechnology, Nature Communications, Advanced Materials, ACS Nano, Nano Letters, Biomaterials等国际权威刊物上发表的论文,被F-1000,福布斯,科学日报,美国化学学会等知名媒体报道。申请PCT 国际专利 2项(1项已转让),中国专利 8 项(1项已转让)。同时,薛向东为《International Journal of Molecular Sciences》编委,《Exploration》首届青年编委,《Biomaterials Translational》和《iRadiology》期刊青年编委,中国药学会生化与生物技术药物专业委员会青年委员,上海市药学会药剂学专委会青年委员(基础药剂学组),中国药学会高级会员,中国生物物理学会会员,中国抗癌协会会员,美国癌症研究协会(AACR)会员;多次在中国暨国际生物物理大会,中国药剂学大会,中国生物材料大会,北加州生物材料大会,美国癌症研究年会上做邀请报告和口头报告。为Nat Biomed Eng, Nat Commun, Adv Mater, ACS Nano, Biomaterials, Adv Funct Mater等学术期刊审稿人。

课题组面向“精准医疗”和人民“大健康”需求,基于纳米技术开发精准诊疗策略与技术。具体包括:

o环境自适应的精准药物递送系统开发

复杂的生物环境会阻碍药物活性成分进入靶细胞发挥药效。通过纳米自组装的方法,构建可在不同生物微环境产生结构、影像信号、表面性质等自适应变化的精准药物递送系统,克服递送屏障并将药物或成像剂精准递送至靶细胞,实现精准的疾病治疗和诊断


o免疫治疗与免疫调控

利用先进的纳米技术对免疫系统进行正向或负向调控,对肿瘤或炎症进行有效的治疗,诊断和预后,并显著提高病人的生存期和生存质量。

摘要:本发明公开了一种亲水药物超分子纳米级给药系统及其组装方法,属 于医药技术领域,将亲水药物,如替拉扎明、替莫唑胺、盐酸吉西他滨、 5-偶氮胞苷或阿霉素等作为模型药物,与带有负电性基团并可提供π-π堆积作用力的疏水性药物,如二氢卟吩,原卟啉,脱镁叶绿酸或 STING 激 动剂等在超声条件下直接共组装,形成纳米级给药系统;该方法不需要化学合成而大大简化了纳米级给药系统的制备。通过实验验证,本发明中超 分子自组装纳米级给药系统在荷瘤小鼠中能有效改善药物的瘤内积累,并能通过协同抗肿瘤作用显著抑制肿瘤的生长和转移。因此,本发明中的超分子组装方法为开发易于制造且多功能的亲水性纳米药物递送系统提供 一个新模式。

实用性及应用性:该方法可通过无化学的绿色手段,将亲水性药物如替莫唑胺、提拉扎明、吉西他滨、5-氮杂胞苷和阿霉素等,自组装形成纳米药物,增加药物的成药性,水溶性和安全性,并提高药效,降低化疗毒副作用

摘要:本发明公开了一种荷电生物分子的普适性递送方法,属于生物分子递送技术领域。本发明公开的方法通过带电荷的大π共轭指导分子,与荷电生物分子等物理混合,在非共价作用力的驱动下直接自组装为一种纳米级递送系统,然后再采用聚乙二醇进行表面化交联,以稳定其自组装结构,并屏蔽表面电荷,以改善纳米粒子的体内药代动力学性质;该方法能够高效、精准地递送荷电生物药物分子,具有良好的稳定性和良好的生物安全性。本发明中的组装方法将为开发易于制造且具有多功能性的纳米药物提供一个新的模式,并指导更广泛的生物分子递送。

实用性及应用性:该方法可通过简单的超分子自组装递送带电荷的生物药物分子,如多肽类药物、信使核糖核酸mRNA、小干扰核糖核酸siRNA及多糖类药物(如透明质酸和壳聚糖等)。可显著增加生物药物分子的药代动力学、生物利用度以及递送效率,增加药效和降低毒副作用。
